These parameters are determined by high-performance liquid chromatography with tandem mass spectrometry.<\/p><p>Plasma concentrations of tolvaptan were measured after ad libitum water intake and ad libitum water withdrawal for 2 h. Plasma samples were then dialyzed at 37degC for 7 h with a rapid equilibrium dialysis 96-well system. Then, aliquots were analyzed using high-performance liquid chromatography with tandem Mass spectrometry.<\/p><p>The pharmacodynamic end points were summarized using descriptive statistics by time point. Results showed that tolvaptan reduced the mean CrCL (creatinine clearance) by four to seven percent in patients with renal impairment. This is comparable to the effects seen in patients with autosomal dominant polycystic kidney disease. However, the reductions were not statistically significant.<\/p><p>Plasma concentrations of tolvaptan showed a rapid onset and were reached at a maximum level about 2 h after dosing. Plasma concentrations decreased in an exponential fashion. The number of tablets prescribed for a given population is not exactly the same across prefectures. The prevalence of tolvaptan in the city may be higher than in the suburbs. Nevertheless, it is still an enigma as to why the number of tolvaptan prescriptions has not increased in recent years.<\/p><p>Although the prevalence of tolvaptan tablets in the city may not have increased in the last few years, the number of prescribed tablets for a given population has definitely increased. Compared with rural areas, urban areas tend to adopt new medicines and treatments first. This study suggests that tolvaptan is a promising addition to the armamentarium.<\/p><p>Tolvaptan is a vasopressin receptor 2 antagonist. It has a number of interesting properties including diuretic actions. It has also been demonstrated that tolvaptan can be effective in treating hyponatremia.
