The estimated bioavailability of s.c. trastuzumab was 77 percent, or 1.45 percent of the reference i.v. regimen (RSE).<\/p><p>In the phase I\/Ib study, the pharmacokinetics of a fixed 600-mg SC dose were compared with those of an IV regimen. The pharmacokinetics of IV and SC regimens were similar. However, the concentrations of the IV regimen were significantly higher. This may be due to the fact that the intravenous regimen was administered in a shorter time period.<\/p><p>Several significant studies have been conducted to assess the bioavailability and preference of subcutaneous and intravenous administration of trastuzumab for early HER2-positive breast cancer.<\/p><p>The study was a multicentre randomised phase III study. Patients were enrolled by their treating physician. Initially, the dose was given in four 1.0 mg injections, which increased over time. Trastuzumab is a standard of care for early HER2-positive breast cancer. In the GAIN-2 study, patients were randomized to either intravenous (IV) or subcutaneous (SC) administration. In the per-protocol set, 30 patients were investigated.
